Özet Root lesion nematodes (RLNs) of genus Pratylenchus are important causes of yield loss in crop products. Pratylenchus thornei (major wheat root-lesion nematode) (RLN) is one of the most important species. P. thornei is an important pathogen of wheat all around the world. In this study was to determine the virulence of Purpureocillium lilacinum (syn: Paecilomyces lilacinus) against P. thornei on wheat in the greenhouse condition. P. thornei is produced on carrot culture using pure culture from Biological Control Research Station in Adana (Turkey). P. lilacinum was subcultured on Potato Dextrose Agar (DifcoTM, Becton Dickinson and Company, USA). The harvested fungal conidia were incorporated in to sterile 0.05% Tween-80 solution and the material was stirred for complete homogeneity. The serial dilutions were prepared and the number of conidia was measured to achieve the 106, 107 and 108 cfu ml-1 concentration under haemocytometer. P. lilacinum were applied at the same time with P. thornei in wheat root [susceptible wheat variety (Seri)]. Approximately 1 ml of P. lilacinus in Tween-80 solution was given by a syringe into the soil. The positive control pots received only water containing P. thornei and negative control posts received just water. For each plants (with 7 replicates), variables that were assessed included, total number of nematodes in both plant root and soil, fresh and dry root weight; fresh and dry weight of the upper parts of plant and plant height. The experiment was harvested in 9 weeks from March to May 2013. The results showed that all concentration of P. lilacinus was reduced P. thornei population. The 108 cfu concentration of fungus improved the fresh and dry root weight. In conclusion, P. lilacinus could be promising control methods against P. thornei in infected area.
